721 Efficacy of silybum marianum extract, manganese PCA and lespedeza capitata extract on hair growth and anchorage in human dermal papilla cells

The hair follicle (HF) undergoes cycles of growth (anagen), regression (catagen) and rest (telogen) phases. Both chronic reactive loss are linked to a dysfunction the HF cycle leading premature loss. Therefore, use specific active ingredients targeting could improve aim this study was evaluate efficacy 3 on anchorage in dermal papilla cells isolated from human (DPC): new patented extract Silybum marianum containing less than 2% silymarin (SME) stimulating shaft by keratin 75 modulation (Patent WO/2021/023820), Manganese PCA (MnPCA) Lespedeza capitata (LCE) WO/2020/020791A1). Antibody array gene reporter assays allowed analyse Receptor tyrosine kinase phosphorylation Wnt/β catenin pathways activation, respectively. ELISA tests were performed quantify release Versican, VEGF DKK1. Activity 5α-reductase (5αR) also measured. SME 30 μg/mL activated factor receptor signaling (EGFR x1.9 PDGFR x2.8 respectively) their downstream effectors (ERK, GSK3, Akt, STAT x1.2 2) after 1h exposure. MnPCA stimulated pathway (+80% at 0.009%) induced both Versican production (x33 0.01%) secretion (x3.3 0.009%). LCE reduced DKK1 (-72% 0.001%) 5αR activity (-59.6% 0.01%). To conclude, modulated DPC acting EGFR/PDGFR pathway. enhanced anagen phase via improved microcirculation VEGF, Finally, favored dihydrotestosterone decrease. Altogether, data suggest that combination + may be useful treatment action anchorage.
